Overview

Tumor necrosis factor ligand superfamily member 5 (TNFSF5), also known as CD40 ligand (CD40L) or CD154, is a key cytokine in the tumor necrosis factor (TNF) superfamily. It plays a central role in immune regulation, particularly in the activation and differentiation of B cells, and is essential for effective immune responses. CD40L binds to its receptor, CD40, on B cells and other antigen-presenting cells, triggering signaling pathways that regulate B cell proliferation, immunoglobulin class switching, and other immune functions. Disruption of the CD40-CD40L pathway impairs humoral immunity and can lead to Hyper-IgM syndrome. Targeting this pathway has been explored for autoimmune diseases.
