Overview
The CD40 ligand pathway, also known as the CD40/CD40L or CD154 pathway, is a critical costimulatory signaling system in the immune response. The interaction occurs between the receptor CD40 and its ligand, CD154 (commonly referred to as CD40L). Engagement of CD40 by its ligand triggers multiple downstream effects including immunoglobulin class switching, memory B cell development, cytokine production, and activation of various signaling pathways such as NFκB, MAPK, and PI3K/Akt. Dysregulation or overactivation is associated with autoimmune diseases, cardiovascular complications, transplant rejection risk, and certain cancers.
