CD40, also known as Cluster of Differentiation 40, is a type I transmembrane protein and a member of the tumor necrosis factor receptor (TNFR) superfamily. It is primarily expressed on antigen-presenting cells (APCs), including B cells, dendritic cells (DCs), macrophages, and monocytes. CD40 is essential for activation of APCs; binding by its ligand CD154 leads to upregulation of costimulatory molecules necessary for effective T-cell priming and differentiation. The central role played by the CD40/CD154 axis makes it a key target for modulating immune responses, and has been implicated in diseases ranging from cancer immunotherapy settings to neurodegenerative disorders like Alzheimer’s disease.
Agonistic anti-CD40 antibodies enhance antitumor immunity by activating APCs. Blockade strategies dampen pathological inflammation or autoimmunity.
