CD44 variant 6 (CD44v6) is a specific isoform of the CD44 cell surface glycoprotein, generated through alternative splicing that includes exon v6. It plays key roles in cell-cell and cell-matrix interactions, adhesion, migration, proliferation, and signaling. Overexpression of CD44v6 is observed in several malignancies, associated with increased invasiveness and metastatic potential. It acts as a co-receptor for receptor tyrosine kinases such as MET, promoting signaling pathways like Ras-MAPK and PI3K/Akt. CD44v6 is a prognostic marker in several cancers and a therapeutic target with monoclonal antibodies and antibody-drug conjugates under development.
