Overview

CD5 antigen-like protein (CD5L), also known as Apoptosis Inhibitor of Macrophage (AIM), is a secreted glycoprotein belonging to the scavenger receptor cysteine-rich (SRCR) superfamily (UniProt, 2024). It primarily functions as a key regulator of immune homeostasis and lipid metabolism, notably by inhibiting fatty acid synthase activity and promoting macrophage survival against apoptotic stimuli (PubMed, 2022). CD5L is significantly involved in the pathogenesis of chronic inflammatory conditions such as atherosclerosis and non-alcoholic fatty liver disease (NAFLD), as well as various malignancies like hepatocellular carcinoma (NIH, 2023). In oncology, the protein has been shown to support tumor cell proliferation and facilitate immune evasion within the tumor microenvironment (PubMed, 2023). Current therapeutic development focuses on utilizing monoclonal antibodies to neutralize its pro-tumorigenic effects or applying recombinant CD5L to address metabolic dysfunction and fibrosis (StatPearls, 2023). Furthermore, CD5L serves as a clinical biomarker for assessing the severity of liver fibrosis and cirrhosis (PubMed, 2021).
