Overview

CD6 (Cluster of Differentiation 6) is a human protein encoded by the CD6 gene. It is a type I glycoprotein with a molecular mass of 105-130 kDa that is expressed primarily on thymocytes, mature T cells, B1a cells, and in various brain regions[1][5][7]. CD6 belongs to the scavenger receptor cysteine-rich (SRCR) superfamily of protein receptors. Its extracellular region contains three SRCR domains, each approximately 100-110 amino acids long[1][5][7]. The X-ray crystal structure of CD6 reveals a nonlinear organization of these consecutive SRCR domains, which is the first such structure reported for consecutive SRCR domains[6][8]. The membrane-proximal SRCR domain (third domain) of CD6 is directly involved in binding to its ligand, CD166/ALCAM (Activated Leukocyte Cell Adhesion Molecule)[5][7]. CD6 is a transmembrane protein with both extracellular and intracellular components, with the intracellular portion involved in signaling pathways[7]. CD6 plays several important roles in immune function: 1. T Cell Activation and Costimulation: CD6 functions as an accessory molecule capable of delivering costimulatory signals to T lymphocytes, contributing to T cell activation and proliferative responses[5][7]. 2. Immunological Synapse Formation: CD6 accumulates at the central part of the mature immunological synapse, where it colocalizes with the TCR/CD3 complex and CD5. It has been implicated in early T cell-APC (antigen-presenting cell) contacts, influencing immunological synapse maturation[5][7]. 3. Thymocyte Development: The CD6-ALCAM interaction has been implicated in thymocyte adhesion and development. CD6-dependent signals contribute to thymocyte survival and positive selection[7]. 4. Pathogen Recognition: CD6 can function as a pattern recognition receptor for microbial components. It can bind to and aggregate several Gram-positive and Gram-negative bacterial strains through recognition of lipoteichoic acid and LPS (lipopolysaccharide), respectively[10]. 5. Anti-apoptotic Function: In B cells, CD6 ligation protects against anti-IgM-mediated apoptosis[5][7]. The primary ligand for CD6 is CD166/ALCAM, which is expressed on a variety of cell types including activated T and B lymphocytes, lymphomas, activated monocytes, thymic epithelium, keratinocytes, melanoma cells, mesenchymal stem cells, neurons, and brain cortex[7]. CD6 also interacts with: - Syntenin-1, a PDZ domain-containing protein that interacts with the cytoplasmic domain of CD6[5] - Bacterial components such as lipoteichoic acid and LPS[10] - TCR/CD3 complex and CD5 on T cells[7] CD6 engagement activates several signaling pathways: - Mitogen-Activated Protein Kinase (MAPK) pathway[7] - CD6 ligation with antibodies can deliver costimulatory signals to T lymphocytes[7] CD6 has been associated with: - Multiple sclerosis susceptibility (certain alleles)[1][2] - Potential role in septic shock syndrome and inflammatory diseases of infectious origin[10] A single nucleotide polymorphism (SNP) in CD6, S351N, found in 10% of genomes analyzed, results in glycosylation that disrupts CD6/CD166 interactions. This variant shows a 10-fold weaker affinity to CD166 compared with the S351 variant[6]. The recombinant soluble form of CD6 (rsCD6) has shown therapeutic potential in animal models. Administration of rsCD6 before lethal LPS challenge significantly improved mice survival, with reduced serum levels of proinflammatory cytokines TNF-α, IL-6, and IL-1β[10]. This suggests potential applications in treating septic shock syndrome or other inflammatory diseases of infectious origin. Three different forms of CD6 have been identified: 1. Membrane CD6 (mCD6): 105-130 kDa, found on cell surfaces 2. Recombinant soluble CD6 (rsCD6): Produced for research and potential therapeutic applications 3. Natural soluble CD6 (nsCD6): A circulating form found in human serum with a molecular mass of approximately 80 kDa[10] This comprehensive information about CD6 highlights its importance in immune function, potential as a therapeutic target, and role in various diseases.

Therapeutic recombinant soluble CD6 (rsCD6) can bind microbial components like LPS and lipoteichoic acid, leading to reduced proinflammatory cytokine production. Antibodies targeting membrane CD6 can deliver costimulatory signals to T lymphocytes or modulate interactions with CD166/ALCAM.
