The CD68 promoter regulatory elements are DNA sequences located upstream of the CD68 gene that govern its high-level, specific expression within the myeloid lineage, particularly in monocytes and macrophages (PMID: 8395535). CD68, also known as macrosialin, is a heavily glycosylated type I transmembrane protein primarily found in the endosomal/lysosomal compartment of mononuclear phagocytes (PMID: 10449303). The regulatory region contains critical binding sites for transcription factors such as PU.1 and C/EBP, which are essential for myeloid-specific gene activation (PMID: 9372915). In the field of gene and cell therapy, the CD68 promoter is frequently employed as a tissue-specific driver to restrict the expression of therapeutic transgenes to macrophages, thereby reducing potential off-target toxicity in other cell types (PMID: 25101542). This approach is particularly relevant for treating lysosomal storage disorders, where macrophages serve as the primary vehicle for enzyme replacement, and in the development of chimeric antigen receptor macrophages (CAR-M) for solid tumor immunotherapy (PMID: 32193531). While highly specific, the use of these regulatory elements can sometimes be limited by lower transcriptional activity compared to constitutive viral promoters, necessitating the use of optimized or synthetic variants to achieve therapeutic protein levels.
The CD68 promoter regulatory elements function as a transcriptional switch that recruits myeloid-specific transcription factors, such as PU.1 and C/EBP, to drive the expression of the CD68 gene or a linked transgene specifically within the mononuclear phagocyte system (PMID: 9372915). In therapeutic applications, these elements are incorporated into viral vectors to ensure that therapeutic proteins are produced only in target myeloid cells, thereby minimizing systemic exposure and off-target effects (PMID: 25101542).
