CD8-positive T cell activation is the process by which naive CD8+ T cells become functionally mature cytotoxic T lymphocytes (CTLs) capable of eliminating infected or malignant cells. This process involves antigen recognition via the TCR, co-stimulation, and cytokine signaling, leading to clonal expansion, metabolic reprogramming, and acquisition of effector functions such as the release of perforin/granzymes, Fas-FasL interaction, and cytokine secretion. CD8+ T cell activation is critical for clearing intracellular pathogens and providing tumor surveillance, but dysregulation can contribute to immunopathology.
Immune checkpoint inhibitors enhance CD8+ T cell activation by blocking inhibitory signals. Cytokines promote proliferation and differentiation. Immunosuppressants suppress activation. Vaccines induce antigen-specific activation. Adoptive cell therapies provide pre-activated and engineered CD8+ T cells.
