Overview

The process by which CD8-positive (cytotoxic) T cells are activated in response to tumor-associated peptides that are presented in complex with the molecular chaperone glycoprotein 96 (gp96). This mechanism is central to certain immunotherapeutic strategies aiming to elicit anti-tumor immune responses. Gp96 interacts with specific surface receptors on APCs, notably CD91, facilitating efficient delivery of its peptide cargo for cross-presentation on MHC class I molecules. The interaction of gp96 with DCs induces their maturation through Toll-like receptor (TLR)-mediated signaling pathways, particularly involving TLR2 and TLR4. The primary outcome is the induction of robust cytotoxic T cell responses against tumor antigens.
