Overview

CD84 is a member of the signaling lymphocyte activation molecule (SLAM) family of immunoglobulin superfamily receptors, expressed primarily on hematopoietic cells of myeloid and lymphoid lineages[2][3][9]. It functions as a homophilic adhesion molecule involved in immune cell interactions and regulates both innate and adaptive immune responses[1][2]. CD84 plays important roles in myeloid-derived suppressor cell (MDSC) development and function, and is notably overexpressed in tumor-infiltrating myeloid cells and acute myeloid leukemia (AML) blasts, supporting tumor progression and immune evasion[2][4][5]. CD84 knockout or inhibition decreases survival and proliferation of AML cells and modulates antioxidant responses via the AKT/NRF2 pathway[4]. Its structural features include extracellular immunoglobulin (IgV and IgC) domains, and it self-associates to form dimers essential for signaling activity[1][9]. No small molecule or approved therapeutic antibody is in clinical use for CD84, but it is recognized as a promising target for antibody-based therapies in oncology and immunology[2][4].
