Cdc2-related kinase 12 (CRK12) is a cyclin-dependent kinase involved in cell cycle regulation and transcriptional control. In humans, the ortholog is CDK12, which phosphorylates the C-terminal domain of RNA polymerase II, regulating transcription of genes involved in DNA repair and genome stability. In protozoan parasites like Leishmania donovani and Trypanosoma brucei, CRK12 is essential for proliferation and survival, making it a potential drug target. Inhibition of CRK12 in parasites leads to cell death.
