Overview

CDC42 effector protein 4 (CDC42EP4) is a member of the CDC42-binding protein family that acts as an effector for Rho GTPases, including CDC42 and TC10, primarily in a GTP-dependent manner[1][3][5]. This protein facilitates actin cytoskeleton organization and induces pseudopodia formation, thereby regulating cell shape and motility[1][3]. CDC42EP4 modulates cytoskeletal architecture in neural and non-neural tissues, being exclusively expressed in Bergmann glia in the cerebellar cortex, where it supports synaptic structure and function by forming complexes that influence glial processes[3]. Its altered expression is linked to dendritic spine deficits observed in schizophrenia and it may also participate in nucleocytoplasmic signaling pathways in neural cells. In non-neoplastic epithelial cells, CDC42EP4 phosphorylation by protein kinase C isoforms modulates its interaction with GTPases and promotes cell migration[3]. There is some evidence for a tumor suppressive role in prostate cancer through regulation of ERK signaling, and it is associated with at least one congenital heart disorder (atrial septal defect 1)[4][5]. There are currently no specific drugs targeting CDC42EP4 or established clinical biomarker/safety data.
