CDC42 effector protein 5 (CDC42EP5) is a binder and downstream effector of CDC42, belonging to the Borg (binder of Rho GTPases) family of proteins. It contains a CRIB (Cdc42/Rac interactive binding) domain that enables binding to active CDC42, thereby modulating its cytoskeleton-organizing functions. CDC42EP5 participates in the regulation of actin filaments, septin organization, cell shape, and the formation of pseudopodia and lamellipodia in fibroblasts. This protein can inhibit c-Jun N-terminal kinase (JNK) independently of CDC42 binding and may participate in the formation of filamentous structures with actin or septins, thereby influencing cell migration, morphology, and possibly cancer-related changes. CDC42EP5 does not act as a classical receptor, enzyme, or ion channel, and there are currently no approved drugs or biomarker applications related to it.
