Overview

CDGSH iron-sulfur domain-containing protein 2 (CISD2) is a membrane-associated member of the NEET protein family characterized by a conserved CDGSH motif that binds a [2Fe-2S] iron-sulfur cluster. CISD2 is located predominantly at the endoplasmic reticulum and mitochondria-associated membranes, where it regulates autophagy, apoptosis, and iron/ROS balance. Its biological roles make it relevant to multiple disease processes, including metabolic disorders, cancer, neurodegenerative diseases, and cardiovascular disease. CISD2 functions by maintaining mitochondrial integrity, antagonizing BECN1-mediated autophagy, and controlling mitochondrial signaling related to cell death and survival. Dysfunction or altered expression of CISD2 has been implicated in aging, neurodegeneration, and susceptibility to metabolic syndromes.
