Overview

CDH13 antisense RNA 2 (CDH13-AS2) is a long non-coding RNA (lncRNA) transcribed from the CDH13 gene locus that regulates the expression of the T-cadherin (CDH13) protein. CDH13-AS2 exerts its regulatory effects by directly binding the 3′ untranslated region (3′-UTR) of CDH13 mRNA, thereby countering the binding of microRNAs such as let-7 that would otherwise destabilize CDH13 transcripts. CRISPR/Cas9 knockout studies show that loss of either CDH13 or CDH13-AS2 in endothelial cells results in similar atherogenic phenotypes, such as suppressed proliferation, migration, and increased apoptosis. This lncRNA is implicated in the stabilization and upregulation of CDH13 expression and, by extension, in the maintenance of vascular health and possibly in cancer and neurodegenerative disease pathways. CDH13-AS2 is regarded as part of a complex non-coding RNA network that fine-tunes CDH13 gene expression across several pathological states but is not a canonical therapeutic target in the sense of a protein, enzyme, receptor, or transporter[1][2][3].
