CDK2-associated and cullin domain-containing protein 1 (CACUL1) is a multifunctional cullin domain protein that regulates cell cycle progression, apoptosis, transcription, and stress responses[1][2][3][4]. CACUL1 promotes cell proliferation by enhancing CDK2 activity at the G1/S transition and is frequently overexpressed in a variety of malignancies, correlating with tumor growth, chemoresistance, and metastasis[1][2][3]. It also functions as a transcriptional modulator—repressing activity of PPARγ and androgen receptor, inhibiting SUMOylation of PML, and modulating Nrf2 ubiquitination through association with the Cul3-Keap1 complex, thereby impacting antioxidant stress response and sensitizing cells to oxidative stress[1][2]. Its expression is subject to regulation by microRNAs in cancer and it is implicated as a potential therapeutic target in cancers and metabolic disorders[1][2][3][4].
