Overview

Cdk5 and Abl enzyme substrate 1 (CABLES1) is a cyclin-dependent kinase binding protein that acts as a tumor suppressor gene, mainly regulating cell cycle progression and apoptosis through interactions with multiple cyclin-dependent kinases (CDK2, CDK3, CDK5) and non-receptor tyrosine kinases including Src, Abl, and Wee1[1][2]. CABLES1 modulates kinase activity, enhancing CDK5 activity (crucial for neuronal development) and inhibiting CDK2-mediated growth via the WEE1 pathway[2]. In addition, it potentiates p53/p73-induced cell death and impacts the Wnt/β-catenin signaling pathway in tumorigenesis; its loss activates β-catenin-dependent transcription and proliferation in cancer models, contributing to tumor progression. CABLES1 is frequently inactivated in human cancers by promoter hypermethylation and loss of heterozygosity, and its deficiency correlates with increased tumor formation, cell proliferation, and delayed cellular senescence[1][2][5].
