CDK5 regulatory subunit-associated protein 2 (CDK5RAP2) is a centrosomal protein that plays a central role in organizing microtubules and coordinating cell division, particularly during neurogenesis. It localizes to the pericentriolar material, facilitates the recruitment and attachment of the γ-tubulin ring complex (γTuRC) to centrosomes, and is involved in regulating spindle formation and centriole cohesion. CDK5RAP2 interacts with CDK5R1 (the regulatory subunit of CDK5), pericentrin, and other centrosomal proteins to maintain centrosomal integrity. Loss-of-function mutations in CDK5RAP2 are linked to autosomal recessive primary microcephaly (MCPH3), characterized by reduced brain size due to impaired neural progenitor proliferation. The protein is also relevant to human brain size variation and has potential links to neurodegenerative disorders such as Alzheimer's disease[1][2][3][4].
