Overview

CDKN2A antisense RNA 1 (CDKN2A-AS1, also known as ANRIL) is a long non-coding RNA implicated in multiple disease contexts, most notably various cancers (including ovarian, lung) and cardiovascular diseases such as atherosclerosis[2]. CDKN2A-AS1 acts in a context-dependent manner—promoting cell proliferation, migration, and invasion by activating the BMP–SMAD signaling pathway through direct binding to SOSTDC1, especially in ovarian cancer[3][4]. In cardiovascular disease, increased CDKN2A-AS1 expression is linked to higher risk and severity of atherosclerosis, likely through effects on vascular inflammation and cell proliferation[2]. In lung adenocarcinoma, elevated CDKN2A-AS1 expression is associated with poor prognosis, supporting its role as a biomarker candidate for patient stratification and prognosis[2]. Its precise mechanisms are still being investigated, but CDKN2A-AS1 may function as a proto-oncogene in multiple tumor types and a regulator of gene expression relevant to disease susceptibility and progression[2][3][4].
