Overview

CDKN2A interacting protein (CDKN2AIP, also known as CARF) is a nuclear RNA-binding protein that regulates the DNA damage response through several signaling mechanisms, notably by modulating p53 and MDM2 activity, influencing cell cycle arrest, senescence, apoptosis, and genomic integrity[1][2]. CDKN2AIP is also involved in spermiogenesis and germ cell development, with essential roles in protamine replacement and nuclear morphology during sperm maturation[3]. Dysregulation or altered expression of CDKN2AIP has been associated with several cancers, infertility, and potentially other disease processes, making it a candidate therapeutic and diagnostic target, as well as a biomarker[1][2][3]. Small molecule inhibitors targeting CDKN2AIP-mediated signaling pathways such as Wnt/β-catenin have been described[1], and ongoing research is expanding its therapeutic relevance.
