Overview

CEA cell adhesion molecule 6 (CEACAM6) is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein and a member of the carcinoembryonic antigen (CEA) family, predominantly involved in intercellular adhesion, tumor cell migration, immune modulation, and is overexpressed in a variety of cancers. It mediates cell-cell and cell-matrix interactions, impacts tumor proliferation, invasion, metastasis, and chemoresistance, and is under investigation as a target for antibody- and cell-based cancer therapies. It also plays roles in neutrophil adhesion and hosts as a receptor for bacterial adhesion (notably in Crohn’s disease)

Mechanism of action

Monoclonal antibodies and CAR-T cells targeting CEACAM6 aim to induce tumor cell death via immune-mediated processes or to block CEACAM6-mediated cell adhesion and downstream signaling pathways (e.g., ERK1/2/MAPK, SRC/FAK/PI3K/AKT)
