Overview

Cell adhesion molecule 1 (CADM1) is a transmembrane glycoprotein and a member of the immunoglobulin superfamily, encoded by the CADM1 gene in humans. It plays a crucial role in mediating cell-cell adhesion, particularly in the nervous system, where it is important for synaptic assembly and maintenance. CADM1 has also been implicated in tumor suppression, with loss of expression observed in several cancers. In metabolic disease, variations near the CADM1 locus are associated with obesity and anorexia, and functional studies suggest CADM1 modulates body weight homeostasis. Mutations in CADM1 have been linked to autism spectrum disorder, implicating it in neurodevelopment. CADM1 interacts with other cell adhesion proteins and influences various intracellular signaling pathways. It is also referred to by several aliases, including SynCAM, TSLC1, SgIGSF, and IGSF4. While CADM1 has not been directly targeted by drugs so far, its roles in cancer and neurodevelopment make it an active area of research. Safety concerns mainly relate to the adverse effects of loss of CADM1 function, including increased cancer susceptibility and neurological consequences.
