Overview

CADM1 antisense RNA 1 (CADM1-AS1) is a long non-coding RNA transcribed in the antisense orientation relative to the *CADM1* gene. It has a tumor suppressive role in several cancers, including renal clear cell carcinoma and hepatocellular carcinoma. Downregulation of CADM1-AS1 is observed in cancer tissues, and low levels are associated with worse patient outcomes. Mechanistically, CADM1-AS1 inhibits cell proliferation, migration, and invasion, causing cell cycle arrest at the G0/G1 phase by modulating AKT/GSK-3β signaling, regulating key cell cycle proteins (cyclins, CDKs, and inhibitors like p15, p21, p27), and promoting apoptosis. Its expression pattern and functional activity make CADM1-AS1 a potentially valuable biomarker and therapeutic target in oncology research. There are currently no drugs specifically targeting CADM1-AS1, and its candidacy as a therapeutic target is under investigation in tumor biology rather than via pharmacological intervention. Biological regulation is primarily via genetic or epigenetic modulation (e.g., siRNA, gain/loss-of-function studies).

Mechanism of action

When overexpressed, suppresses phosphorylation of AKT and GSK-3β by upregulating PTEN. Downregulates cell cycle-associated proteins (cyclin D, cyclin E, CDK2, CDK4, CDK6). Upregulates cell cycle inhibitors (p15, p21, p27). Causes G0/G1 phase cell cycle arrest, inhibiting proliferation. Regulates the “CADM1-AS1/CADM1 mRNA gene pairs” to instantiate tumor suppressive effects.
