Cell cycle and apoptosis regulatory pathways encompass the complex molecular signaling cascades that control cell division and programmed cell death (StatPearls, 2023). These pathways are essential for maintaining genomic integrity and tissue homeostasis by ensuring that only healthy cells proliferate and that damaged cells are eliminated (NIH, 2022). In many pathological states, especially cancer, these pathways are frequently dysregulated through the overexpression of oncogenic drivers like Cyclin D1 or the loss of tumor suppressors like p53 (Nature Reviews Cancer, 2021). Therapeutic strategies often target specific nodes within these pathways, such as using CDK4/6 inhibitors to halt the cell cycle or BCL-2 inhibitors to overcome apoptosis resistance (FDA, 2020). While these interventions are potent in treating malignancies, they often present challenges such as myelosuppression and the development of secondary resistance (PubMed, 2022). Understanding the crosstalk between these two pathways is crucial for developing effective combination therapies that can simultaneously inhibit growth and promote death in target cells (Cell Death & Disease, 2021).
