Cell cycle and proliferative signaling pathways encompass the integrated network of molecular signals that regulate the progression of a cell through growth, DNA replication, and division [1, 7]. This system is primarily governed by cyclin-dependent kinases (CDKs) and their regulatory cyclin partners, which act as essential nodes at various checkpoints (e.g., G1/S and G2/M) to maintain genomic stability [1, 6, 8]. Upstream mitogenic cascades, including the MAPK/ERK and PI3K/AKT/mTOR pathways, relay extracellular growth factor signals to the cell cycle machinery, often by inducing the expression of D-type cyclins [3, 5, 7]. Dysregulation of these pathways is a fundamental hallmark of cancer, where mutations in oncogenes or the loss of tumor suppressors like RB1 and p53 lead to persistent, uncontrolled proliferation [2, 4, 10]. Pharmacological intervention typically targets specific components of this network, such as CDK4/6 inhibitors (e.g., palbociclib) or inhibitors of upstream kinases, to induce cell cycle arrest or apoptosis in malignant cells [1, 2, 5]. However, because these pathways are also active in normal proliferating tissues, therapeutic use is often limited by toxicities such as myelosuppression and gastrointestinal distress [1, 11].
