Overview

The cell cycle progression machinery refers to the network of molecular components that regulate and drive the orderly sequence of events leading to cell division. This machinery ensures accurate DNA replication, segregation of genetic material, and cytokinesis, resulting in two genetically identical daughter cells. The process is tightly regulated to maintain genomic integrity and respond appropriately to internal and external cues. Key components include cyclin-dependent kinases (CDKs), cyclins, ubiquitin ligase complexes (SCF and APC/C), retinoblastoma protein (Rb), and E2F transcription factors. Dysfunction or deregulation contributes directly to tumorigenesis. Many cancers harbor mutations affecting core components such as pRb pathway elements or overexpressing certain cyclins/CDKs.
