Cyclin D1 is a key cell cycle regulatory protein that controls the G1-S phase transition. It forms complexes with CDK4/CDK6, phosphorylating RB1 and promoting E2F-mediated transcription of genes required for DNA replication. Cyclin D1 expression is regulated by mitogenic signals and its degradation is essential for proper cell cycle control. Beyond its role with CDKs, Cyclin D1 has CDK-independent functions including modulation of transcription factor activity and regulation of cellular metabolism. Overexpression of cyclin D1 contributes significantly to tumorigenesis and is a potential biomarker in several cancers.
