Overview

Cell death-inducing p53-target protein 1 (CDIP1) is a monotopic membrane protein involved in regulating cell death. Initially identified as a pro-apoptotic target of p53, CDIP1 reinforces apoptotic signaling by activating the extrinsic cell death pathway, including caspase-8 cleavage and TNF-alpha induction, making cells highly sensitive to TNF-alpha-mediated apoptosis. CDIP1 shares membrane topology with LITAF and participates in membrane-associated signaling events. It serves as an alternative host receptor for the Bacillus cereus hemolysin BL toxin, linking it to mediation of bacterial toxicity and inflammation. In cardiovascular biology, CDIP1 activity is regulated by noncoding RNA and microRNAs, affecting hypoxia/reoxygenation-induced injury. In cancer, its expression modulates apoptotic responses, notably in non-small cell lung cancer where IL-33-induced miR-128-3p can inhibit CDIP1. CDIP1 integrates p53-mediated intrinsic signals with death receptor-mediated extrinsic pathways, and its versatile roles make it a potential therapeutic target in cancer, cardiovascular injury, and infectious diseases
