Overview

Cell division control protein 45 homolog (CDC45) is an essential eukaryotic protein required for the initiation and elongation phases of chromosomal DNA replication. In humans, CDC45 is a core component of the CMG helicase complex (composed of CDC45, MCM2-7, and GINS proteins), where it is indispensable for the assembly and activation of the replicative helicase machinery. The protein plays a pivotal role in the loading of DNA polymerase α onto chromatin and interacts with numerous DNA replication factors, including members of the MCM complex and origin recognition complex (ORC) proteins. CDC45's biological importance is highlighted by its highly conserved function from yeast to humans and its critical involvement during S-phase checkpoint response to DNA damage. CDC45 has no known intrinsic enzymatic activity and acts through physical association within multi-protein assemblies. Mutations or deregulation of CDC45 are associated with impaired DNA replication, cancer, and developmental disorders such as Meier-Gorlin syndrome
