Overview

Cell division cycle 7-related protein kinase (CDC7) is a serine/threonine protein kinase essential for the initiation of DNA replication during the cell cycle. It phosphorylates components of the minichromosome maintenance (MCM) complex, allowing DNA replication to begin. CDC7's overexpression is linked to tumorigenesis, making it a therapeutic target in oncology. Inhibiting CDC7 impairs cancer cells' ability to respond to replication stress, leading to DNA damage and apoptosis. Several inhibitors, such as SGR-2921, are under clinical investigation. CDC7 expression peaks functionally during S phase and is regulated post-transcriptionally by microRNAs. The CDC7 gene is highly conserved across eukaryotes.
