Overview

Cell division cycle associated 1 (CDCA1), also known as NUF2, is a critical protein component of the NDC80 kinetochore complex, which facilitates the stable attachment of spindle microtubules to kinetochores during mitosis [1]. While CDCA1 expression is primarily restricted to the testis in healthy adults, it is significantly upregulated in a wide variety of malignancies, including non-small cell lung cancer, bladder cancer, and colorectal cancer [2]. The CDCA1-derived peptide, specifically the 9-mer epitope CDCA1-63 (sequence: VYGFVRACL), is processed and presented on the cell surface by the Human Leukocyte Antigen (HLA)-A*24:02 molecule [3]. This peptide-MHC complex serves as a specific target for the T-cell receptor (TCR) of cytotoxic T lymphocytes (CTLs), which can recognize and selectively eliminate the tumor cells [4]. Therapeutic strategies targeting this complex include peptide-based vaccines, such as OTSGC-A24, and TCR-engineered T-cell therapies, which aim to induce a robust anti-tumor immune response in patients carrying the HLA-A*24:02 allele [5].
