Overview

The HLA-A24–presented CDCA1 peptide is a molecular complex comprising a peptide derived from the Cell Division Cycle Associated 1 (CDCA1) protein, also known as NUF2, bound to the Human Leukocyte Antigen A*24:02 (HLA-A*24:02) molecule (PubMed: 19509139). CDCA1 is a critical component of the Ndc80 kinetochore complex, playing a vital role in chromosome segregation during mitosis (UniProt: O75935). While its expression is restricted in normal tissues, it is highly overexpressed in various cancers, including non-small cell lung cancer and gastric cancer, functioning as a tumor-associated antigen (PubMed: 21107155). The presentation of the CDCA1-63 peptide (VYGIRLEHF) by HLA-A*24:02 on the tumor cell surface serves as a target for cytotoxic T lymphocytes (CTLs), which can be induced through vaccination (PubMed: 24331138). Therapeutic development has focused on peptide vaccines, such as the CDCA1-63 peptide, to stimulate an immune response specifically against cells displaying this complex. These vaccines are often evaluated in patients with HLA-A*24:02-positive status, a common allele in East Asian populations, to ensure target recognition and efficacy (PubMed: 27503017).
