Overview

Cell division cycle-associated protein 8 (CDCA8), also known as Borealin, is a core component of the chromosomal passenger complex (CPC), along with Aurora B kinase, INCENP, and Survivin[1][3][5]. The CPC is essential for accurate chromosome segregation, spindle formation, stabilization of the bipolar mitotic spindle, and proper cytokinesis during mitosis[1][2][3][5]. CDCA8 plays a crucial role in maintaining CPC integrity and ensuring cell-cycle progression, and its function is required for chromatin-induced microtubule stabilization. CDCA8 is overexpressed in several cancers, especially lung cancer, where it is frequently co-activated with Aurora kinase B; this overexpression correlates strongly with poor patient outcomes[1]. Aurora B kinase phosphorylates CDCA8, stabilizing the protein in cancer cells. Experimental inhibitors targeting the CDCA8-Aurora B interaction and RNA interference techniques suppress tumor cell proliferation, highlighting CDCA8 as a promising anti-mitotic cancer therapeutic target[1][5]. CDCA8 interacts directly with INCENP, Survivin, and Aurora B kinase to execute its functions[2][3][5].
