Cell division cycle protein 123 (CDC123) is an ATP-dependent chaperone critical for assembling the eukaryotic translation initiation factor 2 (eIF2) complex, thereby enabling the onset of protein synthesis. It binds specifically to the gamma subunit of eIF2, supporting its assembly and function. CDC123's regulation is tightly integrated with cellular proliferation and cell cycle control, and aberrant expression or stability of CDC123 (regulated in part by the deubiquitinase USP9X) promotes tumorigenesis, especially in breast cancer. As an essential protein for normal cell growth, CDC123 represents a potentially targetable molecule in oncology and disease contexts, but its indispensable role in the basic machinery of protein biosynthesis raises significant safety considerations.
