Overview

Cell division cycle protein 16 homolog (CDC16) is a core component of the anaphase-promoting complex/cyclosome (APC/C), a multiprotein E3 ubiquitin ligase complex essential for regulation of cell division. The APC/C complex mediates ubiquitination and targeted degradation of key cell cycle proteins, primarily cyclins, to facilitate progression through mitosis and entry into the G1 phase. CDC16 contains tetratricopeptide repeat (TPR) domains, which are important for protein-protein interactions and assembly of the complex. CDC16 and associated subunits (such as CDC23 and CDC27) are highly conserved across eukaryotes and play a pivotal role in mitotic regulation, ensuring proper timing of chromosome segregation and cell division[1][2][7]. Defects or dysfunctions in CDC16 and the APC complex are associated with abnormal cell proliferation and can contribute to cancer development. Mutations are also linked to certain forms of hereditary deafness and may be implicated in additional pathological processes[2]. Note: There are currently no well-established drugs, biomarkers, mechanisms of action, or safety concerns directly targeting CDC16, reflecting its fundamental and ubiquitous role in cell division and the lack of specific therapeutic agents[2].
