Overview

Cell division cycle protein 20 homolog B (CDC20B) is a WD40-repeat–containing protein required for centriole release during multiciliated cell differentiation. Unlike the canonical CDC20, which is an activator of the anaphase-promoting complex (APC/C) and a well-known cell cycle regulator, CDC20B lacks the domains necessary for APC/C binding and activation. Instead, it functions in association with deuterosomes and is essential for separating newly formed centrioles from these structures, enabling subsequent cilia production in vertebrate multiciliated cells. CDC20B is specifically expressed during the phase of centriole amplification in maturing MCCs and forms complexes with proteins such as PLK1 and SPAG5 to coordinate the local activation of Separase, the protease necessary for centriole disengagement. Disruption of CDC20B impairs ciliogenesis, highlighting its unique and essential role in multiciliary tissue biology.
