Cell growth-regulating nucleolar protein Ly1 antibody reactive (LYAR) is a zinc finger nucleolar protein that functions as a transcription factor with DNA-binding activity. It regulates genes involved in cell proliferation, ribosome biogenesis, erythrocyte development, and immune response. LYAR is highly expressed in embryonic stem cells, where it sustains cell self-renewal and controls differentiation via nucleolin stabilization and rRNA processing. In cancer, LYAR is frequently overexpressed, promoting tumor progression, metastasis, and poor clinical outcomes by enhancing cell migration, invasion, and suppressing oxidative stress response genes. LYAR also negatively regulates the innate antiviral response and pro-inflammatory cytokine transcription by interfering with IRF3 and NF-κB activity. Its clinical relevance as a prognostic biomarker is supported across several tumor types, but no pharmacological agents currently target LYAR directly.
