Overview

Cell membrane and protein surface refer to the fundamental structural and functional interfaces of biological systems rather than discrete molecular targets. The cell membrane is a dynamic lipid bilayer that provides compartmentalization, regulates molecular transport, and serves as a scaffold for signaling complexes (Alberts et al., 2002). Protein surfaces are the specific topographies where molecular recognition occurs, facilitating the protein-protein interactions (PPIs) essential for almost all cellular processes (Wells & McClendon, 2007). While certain therapeutic agents, such as polymyxin antibiotics, target bacterial membranes or small molecules are developed to bind protein-protein interfaces, these structures are generally too broad to be considered individual therapeutic targets (Nation et al., 2015). In drug discovery, they are typically viewed as the physical context or site of action rather than a discrete molecular entity like a specific receptor or enzyme. Consequently, this designation is considered incorrect for a specific target profile due to its lack of molecular specificity and its inclusion of vast, heterogeneous biological components.
