Cell membrane phospholipid synthesis refers to the biochemical pathways by which cells produce phospholipids, the primary structural components of biological membranes. These amphipathic molecules form lipid bilayers that are essential for cell compartmentalization, signaling, and membrane protein function. Most phospholipid synthesis occurs on the cytosolic face of the endoplasmic reticulum (ER). The process begins with glycerol-3-phosphate and fatty acyl-CoA molecules. Key regulatory enzymes include GPAT/LPAAT, CCT, and PSD. Phospholipid composition is dynamically remodeled after initial synthesis through actions of various phospholipases. Lipid transport proteins facilitate movement between organelles. Phospholipid biosynthesis is essential for membrane biogenesis, maintenance of organelle identity and signal transduction pathways.
