Overview

Cell-surface D-galactose-containing glycoconjugates are a diverse class of glycoproteins and glycolipids characterized by the presence of terminal or internal D-galactose moieties (Varki et al., 2015). These carbohydrates are integral components of the glycocalyx and play essential roles in biological processes such as cell-cell communication, adhesion, and the regulation of receptor signaling (Gabius, 2006). In various diseases, particularly oncology, the expression and branching of these galactose-containing structures are often altered, leading to the presentation of tumor-associated carbohydrate antigens (Pinho & Reis, 2015). These residues serve as the primary docking sites for several potent biological agents, including Type II ribosome-inactivating proteins (RIPs) like viscumin (from mistletoe) and ricin (Lord et al., 2003). Upon binding to these galactose targets, the agents are internalized via endocytosis, where they can exert cytotoxic effects by halting protein synthesis (Stirpe, 2004). Consequently, these glycoconjugates are significant targets in the development of lectin-based therapies and targeted drug delivery systems (Bies et al., 2004).
