Overview

The Cell surface glycoprotein CD200 receptor (CD200R) family consists of several transmembrane proteins belonging to the immunoglobulin superfamily that modulate immune cell activation (UniProt Q8TD46). The primary member, CD200R1, is an inhibitory receptor expressed on myeloid cells and lymphocytes; its interaction with the CD200 ligand delivers a signal that suppresses inflammatory responses and maintains immune homeostasis (PubMed: 12692555). While CD200R1 contains an inhibitory signaling motif, its paralogs—such as CD200R2, CD200R3, and CD200R4 in rodents—often function as activating receptors by pairing with the DAP12 adapter protein (PubMed: 15864271). In cancer, the CD200-CD200R1 pathway is frequently upregulated, allowing tumors to evade the immune system, which has led to the development of CD200R1-targeted antagonists to enhance anti-tumor immunity. Conversely, CD200R1 agonists are being explored as therapeutic agents for autoimmune and chronic inflammatory diseases to reduce tissue damage caused by overactive immune cells (PubMed: 21169541).
