Overview

CD200 receptor (CD200R), also known as OX2 receptor, is a cell surface transmembrane glycoprotein primarily expressed on myeloid lineage cells—including macrophages, dendritic cells, neutrophils, and mast cells—as well as some lymphoid populations such as CD4+ T cells. Its main ligand is CD200 (OX2), a widely distributed membrane glycoprotein. Engagement of CD200R by its ligand leads to potent inhibition of immune activation.

Mechanism of action

Engagement of CD200R by CD200 leads to phosphorylation at NPXY motifs which recruit DOK adaptor proteins, primarily DOK2, leading to downstream Ras/MAPK pathway inhibition and subsequent immune suppression.
