Cell wall biogenesis 43 C-terminal homolog (CWH43) is a human enzyme localized to the endoplasmic reticulum and cell membrane, involved in lipid remodeling during the maturation of GPI-anchored proteins. It acts upstream of processes such as protein localization, and N-linked glycosylation, contributing to membrane protein trafficking and compartmentalization. CWH43 has been implicated as a tumor suppressor gene in colorectal cancer and can serve as a prognostic marker of disease progression. Protein coding defects in CWH43 are associated with rare diseases such as communicating hydrocephalus and Seckel syndrome 8. Its role is essential for proper protein anchoring and membrane structure, and dysfunction can lead to diverse developmental and oncologic pathologies.
For potential therapeutics targeting CWH43, postulated mechanisms may include: Modulation of lipid remodeling or GPI anchor maturation to affect membrane protein trafficking; Restoration or inhibition of CWH43 enzymatic activity in disease models (particularly cancer).
