Overview

Cellular DNA serves as the primary repository of genetic information and is a critical therapeutic target, particularly in oncology. Drugs targeting DNA typically function by disrupting its structure or function, thereby inhibiting replication and transcription, which leads to cell cycle arrest and apoptosis (NIH, 2024). This category also encompasses "other biomolecules" such as RNA and cellular proteins that may be non-specifically modified by highly reactive agents like alkylators or platinum-based compounds (PubMed, 17013861). While effective at killing rapidly dividing cancer cells, these interactions are often non-selective, leading to significant side effects and potential long-term risks like secondary malignancies (StatPearls, 2023). Understanding the interaction between drugs and these fundamental cellular components is essential for developing both traditional chemotherapies and targeted DNA-damaging agents.

Mechanism of action

Drugs targeting cellular DNA and other biomolecules primarily act through covalent binding (alkylation), cross-linking of DNA strands, intercalation between base pairs, or the induction of single- and double-strand breaks, which collectively inhibit DNA synthesis and cellular replication (StatPearls, 2023; PubMed, 17013861).
