Cellular inflammatory signaling pathways are complex networks of biochemical cascades that coordinate the body's response to harmful stimuli, such as pathogens, tissue injury, or environmental stressors [Oncotarget, 2023; NIH, 2023]. These pathways, including the NF-κB, MAPK, and JAK-STAT cascades, integrate signals from various cell-surface receptors like Toll-like receptors (TLRs) and cytokine receptors to trigger the expression of pro-inflammatory genes [OmicsOnline, 2024; Cell Signaling Technology]. While essential for host defense and tissue repair, chronic or dysregulated activation of these pathways is a hallmark of numerous diseases, including rheumatoid arthritis, inflammatory bowel disease, and various cancers [Oncotarget, 2023; SciTechnol, 2023]. Pharmacological intervention typically involves targeting specific components within these pathways, such as kinases or cytokines, to restore immune homeostasis and alleviate disease symptoms [OmicsOnline, 2024; NIH, 2021]. Furthermore, the crosstalk between different signaling networks, such as the interaction between the Hippo and mTOR pathways, adds another layer of complexity to the inflammatory response [MDPI, 2023]. However, the broad involvement of these pathways in normal cellular functions presents significant challenges for achieving therapeutic selectivity and avoiding adverse effects like immunosuppression [AssayGenie, 2024; Longdom, 2023].
Drugs targeting these pathways typically inhibit specific molecular nodes within the cascades, such as cytokine receptors (e.g., TNFR, IL-6R), intracellular kinases (e.g., JAK, MAPK, IKK), or transcription factors (e.g., NF-κB), thereby modulating the production and activity of pro-inflammatory mediators [OmicsOnline, 2024; NIH, 2021; Longdom, 2023].
