Overview

The cellular iron handling system is a coordinated network of proteins and regulatory elements that maintain intracellular iron levels within a narrow physiological range. Key components include Transferrin Receptor 1 (TfR1) for iron uptake, Ferritin for storage, and Ferroportin for export, all of which are post-transcriptionally regulated by Iron Regulatory Proteins (IRP1 and IRP2) through Iron-Responsive Elements (IREs) (NIH, 2013). Systemically, the hormone hepcidin acts as a master regulator by inducing the degradation of ferroportin to limit iron release into the plasma (NIH, 2019). Dysregulation of this system is implicated in various pathologies, including hereditary hemochromatosis, iron-deficiency anemia, and cancer, where increased iron demand supports rapid proliferation (RSC, 2013). Therapeutic strategies targeting this system include iron chelators to treat overload, hepcidin mimetics for iron-loading anemias, and TfR1-targeted delivery systems for oncology (NIH, 2013).
