Organelles like the mitochondria and endoplasmic reticulum provide the structural framework for biochemical reactions, while biomolecules including proteins, nucleic acids, and lipids serve as the functional actors within these compartments [2]. Although these components are the sites where drug interactions occur, the term itself refers to a broad biological category rather than a specific, druggable molecular target [3]. Dysregulation of these systems is a hallmark of numerous pathologies, including cancer, where organelle stress and metabolic reprogramming are common [4]. Pharmacological agents generally target specific receptors or enzymes located within these structures, such as the targeting of lysosomal enzymes in enzyme replacement therapies [5]. Consequently, while vital to pharmacology, this classification is too heterogeneous to facilitate targeted drug design in its collective form.
