Overview

Cellular phospholipid membranes and lipid rafts are dynamic, organized structures that form the fundamental boundary of the cell and its internal compartments. Lipid rafts are specialized microdomains enriched in cholesterol and sphingolipids that act as scaffolding platforms for various signaling proteins, including G protein-coupled receptors and tyrosine kinases (Simons & Toomre, 2000, Nature Reviews Molecular Cell Biology). These structures play critical roles in biological processes such as signal transduction, endocytosis, and viral entry/exit (Lingwood & Simons, 2010, Science). In disease states, lipid rafts can facilitate the hyperactivation of oncogenic signaling or serve as entry points for pathogens like HIV and influenza (Mollinedo & Gajate, 2015, Drug Resistance Updates). Therapeutic strategies targeting these domains include the use of alkylphospholipids to induce apoptosis in cancer cells or polyene antibiotics that disrupt fungal membrane integrity by binding to specific sterols (Sezgin et al., 2017, Nature Reviews Molecular Cell Biology). Because these structures are ubiquitous, a major challenge in drug development is achieving selectivity for diseased versus healthy membranes to minimize systemic toxicity (Pike, 2006, Journal of Lipid Research).
