Overview

Cellular repressor of E1A-stimulated genes 2 (CREG2) is a glycoprotein primarily expressed in the brain and shares homology with CREG1, which is better studied. CREG2 is structurally related to oxidoreductases but, like CREG1, lacks enzymatic function due to inability to bind the cofactor flavin mononucleotide[1][2]. It appears to localize to the Golgi apparatus, endoplasmic reticulum, and extracellular space, and may also localize to the endocytic-lysosomal system[3]. The broader CREG family antagonizes cell proliferation and promotes differentiation and senescence, though direct mechanistic studies on CREG2 are scarce and most functional data are extrapolated from CREG1[1][2]. CREG2 may play a vital role in brain development and normal brain physiology[1][2]. There is currently no evidence that CREG2 is a recognized therapeutic target with known drugs or that it serves as a biomarker or poses special safety challenges. Its biological significance is suggested by its conservation, tissue-specific expression, and potential involvement in the regulation of growth and differentiation in the brain[1][2][3]. CREG2 is not classified as a receptor, transporter, channel, or enzyme, but rather as a secreted or lysosomal glycoprotein with regulatory roles in cell biology[1][2][4]. Data specific to diseases or pharmacological targeting of CREG2 are lacking as of now.
