The vascular niche is a specialized microenvironment composed of endothelial cells, perivascular cells (such as pericytes and mesenchymal stem cells), and extracellular matrix components that collectively regulate stem cell maintenance, proliferation, and differentiation (Butler et al., 2010, Nature Reviews Cancer). It serves as a critical site for the maintenance of hematopoietic stem cells in the bone marrow and neural stem cells in the brain, providing essential paracrine signals like CXCL12 and SCF (Pinho & Frenette, 2019, Blood). In pathological states, particularly oncology, the vascular niche is often remodeled to support tumor growth, facilitate metastasis, and provide a protective sanctuary for cancer stem cells against therapeutic intervention (Hira et al., 2017, Scientific Reports). Therapeutic strategies targeting this niche involve agents that inhibit pro-angiogenic factors like VEGF (e.g., bevacizumab) or disrupt the interaction between stem cells and the niche using CXCR4 antagonists (Ferrara & Adamis, 2016, Nature Reviews Drug Discovery). Because it represents a complex system of cellular and molecular interactions rather than a single protein, it is considered a therapeutic concept or anatomical site rather than a discrete molecular target.
Drugs targeting the vascular niche primarily function by inhibiting pro-angiogenic signaling (e.g., VEGF/VEGFR blockade), disrupting the physical and chemical interactions between stem cells and their perivascular environment, or modulating the paracrine output of endothelial cells to inhibit tumor progression (Ferrara & Adamis, 2016, Nature Reviews Drug Discovery; Rafii et al., 2016, Nature).
